Why work backwards
The usual direction gives you whatever number falls out. Often that is something like 13.5 units on a barrel printed in whole units, which means every dose from that vial is an approximation, in the same direction, for as long as the vial lasts.
Since the diluent volume is a free choice that does not affect how much peptide you get, there is no reason to accept an awkward figure. Pick the mark you want to draw to and add the volume that puts the dose exactly there. The main calculator runs it the other way once the vial is already mixed.

Every row delivers the identical 500 mcg and the identical number of doses from the vial. The only thing that changes is how much liquid carries it and what you read on the barrel. Larger targets need more diluent, which the vial has to physically hold, so the practical ceiling is the vial rather than the arithmetic.
Questions
- Does the diluent volume change how much peptide I get?
- No. It changes the concentration and therefore the volume you draw, but the amount of peptide in that draw is the same and the number of doses in the vial is unchanged. Only the reading on the syringe moves.
- Why aim for a round number of units?
- Because a barrel is printed in whole units, and a figure that lands between two marks has to be approximated every single time. Choosing the diluent so the dose falls on a printed mark removes that error from every subsequent dose.
- Is there a maximum amount of water I can add?
- The physical limit is what the vial holds, which is usually 2 to 5 mL for a small vial with a good deal of headroom. Beyond that the vial simply will not take it, so check the volume against the vial before drawing it up.
