How to Reconstitute a Peptide
Eight steps, of which two get rushed: adding the diluent gently, and labelling the vial afterwards.
The steps
Work out the diluent volume first
Decide it before anything is opened, because it is the one choice that is awkward to change afterwards. The volume sets what one syringe unit will be worth, so pick the one that puts your dose on a printed mark.
Let the vial reach room temperature
A vial taken straight from the fridge has cold glass and condensation on it. Letting it settle first avoids both.
Wipe both stoppers
The peptide vial and the diluent vial. An alcohol swab on each, and let them dry rather than wiping them off wet.
Draw the diluent
Draw the volume you calculated. A syringe larger than an insulin syringe is easier here, since you are measuring millilitres rather than units.
Add it down the wall of the vial, slowly
Angle the needle so the stream runs down the inside wall rather than falling directly onto the powder. Peptides are fragile and a jet straight into the pellet is the part of this process most likely to damage them.
Let it dissolve. Do not shake
Most of it goes into solution on its own within a few minutes. If it needs help, roll the vial gently between your fingers or swirl it. Shaking denatures peptides and produces foam that makes accurate drawing harder.
Look at it
It should be clear and free of particles. Cloudiness, visible bits or discoloration mean something is wrong, and the label for the diluent says the same about any reconstituted solution.
Label the vial
Write the date you mixed it, what is in it, and the concentration. The date is the one that matters later, because nothing on the vial records when it was first entered.
The two that get rushed
Adding the diluent too fast. It takes a few extra seconds to angle the needle at the wall and let the stream run down it, and it is the single step where the peptide can be damaged before it has dissolved.
Not labelling the vial. Nothing on a reconstituted vial records the concentration you chose or the day you first entered it, and both matter later. The concentration is what lets you re-derive any dose without opening the calculator again; the date is what the in-use period counts from.

Questions
- Can you shake a peptide vial to dissolve it?
- No. Shaking can denature the peptide and introduces foam that makes drawing an accurate volume harder. Gentle swirling or rolling the vial between your fingers achieves the same thing without either problem.
- Why add the water down the side of the vial?
- So the stream does not hit the lyophilised pellet directly. Adding it down the wall lets it reach the powder gradually, which is gentler on the peptide.
- How long does a peptide take to dissolve?
- Usually a few minutes with occasional gentle swirling. If a vial has not dissolved after a reasonable time, something is unusual about it and forcing it with agitation is not the answer.
- What if the solution is cloudy?
- Do not use it. The diluent label instructs inspecting reconstituted solutions for clarity and absence of precipitation or discoloration before administration, and cloudiness fails that check.
